DMT is an active ingredient extracted from multiple precious herbal materials. It effectively inhibits *Cutibacterium acnes* and mites, and also shows good inhibitory activity against *Candida albicans*, *Staphylococcus aureus* and other microorganisms. At an addition level of 2%, it achieves 100% mite inhibition rate within 24 h under test conditions

Mechanism of Action

1. Demodex regulation pathway: Act on cell membrane structure of Demodex, reduce skin Demodex load and relieve redness and skin bumps associated with Demodex.
2. Microbiome balancing pathway: Affect cell structure of *Cutibacterium acnes* and *Malassezia*; inhibit microbial biofilm formation, reduce proliferation of harmful microorganisms in hair follicles and improve inflammatory skin bumps.
3. Sebum regulation pathway: Inhibit type Ⅱ 5α-reductase activity, reduce conversion of testosterone into dihydrotestosterone (DHT), lower excessive sebum secretion, improve oily skin and mitigate the trigger of pore clogging.
4. Soothing pathway: Downregulate expression of inflammatory mediators IL‑1β, IL‑6, TNF‑α and PGE₂, relieve skin inflammatory stress induced by UVB and lipopolysaccharide, and alleviate red papules.
5. Antioxidant pathway: Scavenge UVB-induced cellular ROS, reduce oxidative stress damage and ease secondary oxidative damage caused by blemishes.
6. Barrier repair pathway: Increase ceramide content in stratum corneum, reduce transepidermal water loss (TEWL), strengthen skin water retention and repair damaged epidermal barrier of blemish-prone skin.

Experimental Data

1. In-vitro mite-killing test: 2% DMT extract achieves total mite mortality within 10 h with 100% mite reduction at 48 h. 2% DMT cream delivers 100% Demodex mortality in 1 h. For human subjects, average Demodex reduction rate reaches 83.27%, and the best reduction for folliculitis samples hits 86.36%.
2. Bacterial biofilm inhibition test: 1000 mg/L DMT achieves 96.3% inhibition against Staphylococcus aureus biofilm.
3. 5α-reductase inhibition test: 2% DMT exhibits 95% inhibitory activity against type II 5α-reductase and reduces DHT production.
4. Cellular anti-inflammation test: At 6 μg/mL DMT, IL‑1β mRNA expression drops by 52.9% and IL‑6 mRNA by 44.4%. At 60 μg/mL, IL‑6 is reduced by approx. 67.11% and TNF‑α by approx. 56.49%. It suppresses PGE₂ production in a dose-dependent manner with maximum inhibition of 75%.
5. Antioxidant ROS scavenging test: 6 μg/mL DMT scavenges 88.9% of UVB-induced ROS in HaCaT cells; 100 μg/mL reduces neutrophil ROS generation by 72%.
6. Skin barrier repair test: After 4 days of application with 2% DMT, TEWL decreases, water retention capacity improves by 1.5 times, and ceramide level in stratum corneum rises by 89%.
