Pyran Brightening Agent
Pyran Brightening Agent
Lipophilic raw material; scavenges both oxygen free radicals (ROS) and nitrogen free radicals (RNS); excellent formulation stability; non-mutagenic, no cytotoxicity and no phototoxicity at the tested concentrations.
Product overview
It is specially designed for skin brightening applications with no cytotoxicity, irritation or sensitization reactions. It delivers remarkable skin whitening benefits and fights photoaging
Composition Information

Mechanism of Action
1. Inhibit tyrosinase activity, block the conversion of tyrosine to dopa and dopaquinone, and reduce pigment biosynthesis.
2. Dual free radical scavenging: Eliminate reactive oxygen species (ROS) and reactive nitrogen species (RNS), relieve skin oxidative stress and mitigate damage induced by UV radiation and external pollutants.
3. Photoprotective effect: Reduce UV-induced damage to human epidermal keratinocytes and lower pigment deposition triggered by photodamage.
Experimental Data
1. In vitro mushroom tyrosinase inhibition test (0.1 mM): Pyran brightening agent achieves 37% tyrosinase inhibition.
2. In vitro human tyrosinase inhibition test (0.1 mM): Pyran brightening agent achieves 43% tyrosinase inhibition.
3. In vitro melanin test on human melanocytes: Treatment with 0.1 mM pyran brightening agent for 5 days yields 45.87% pigment lightening improvement; 9-day treatment yields 61.2% pigment lightening improvement. The brightening effect is dose-dependent.
4. Keratinocyte photoprotection test: After pretreatment with pyran brightening agent, cell viability under UV irradiation is significantly improved. Cell proliferation capacity increases by 190.4% at 47 μg/mL and 254.88% at 150 μg/mL.
5. Human facial skin brightening test (0.1% inclusion, twice daily for 2 months, n=20): Significant improvement in skin luminance and brightening index is observed at day 30 and day 60 compared with blank control.
6. Human pigment spot test (0.5% inclusion): After 30 days of application, melasma appearance improves by 48% and freckle appearance improves by 47%. At day 60, melasma appearance improves by 72% and freckle appearance improves by 67%.

Key benefits
1. Excellent formulation stability; for oil-in-water emulsion stored at room temperature and 40℃ for 6 months, the active content shows no obvious decline under different pH conditions.
2. Multi-pathway action: inhibits tyrosinase to reduce melanin formation, scavenges ROS/RNS free radicals and delivers photoprotective effect.
3. Favourable safety profile: negative in Ames test (no mutagenicity); no eye irritation, no photodamage; no cytotoxicity observed in tests of keratinocytes, fibroblasts and melanocytes.
4. Effective at low addition levels; both in vitro cell assays and human tests confirm whitening and spot-fading effects, with improvements observed on chloasma and freckles.
Applications and formulation
