SaxiMimic Peptide

Wrinkle-Relax Peptide mimics saxitoxin, one of the most toxic known marine biotoxins and a major toxin responsible for paralytic shellfish poisoning. It binds to the toxin receptor on Na⁺ channel proteins of neurons and sarcolemma, blocking Na⁺ channels and preventing sodium influx. This disturbs neuromuscular transmission and induces relaxation and paralysis of voluntary muscles, thereby delivering instant wrinkle reduction

Mechanism of Action

1. Dynamic wrinkle soothing pathway: Saxitoxin-like polypeptide binds to Na⁺ channel receptors on neurons and sarcolemma, blocks Na⁺ influx, interferes with neuromuscular signal transmission, relaxes expression-related muscle tension and improves dynamic expression lines.
2. Mitochondrial anti-aging pathway (ergothioneine + NAD⁺): Ergothioneine enters cells via OCTN‑1 transporter and scavenges intracellular and extracellular free radicals. It enhances cGPDH activity to facilitate NAD⁺ production. NAD⁺ activates Sirt1 deacetylase, regulates mitophagy, assists DNA repair, stabilizes telomeres, maintains mitochondrial homeostasis and delays cellular senescence.
3. Soothing pathway (Zanthoxylum armatum extract): Reduce the production of inflammatory factors, scavenge free radicals and mitigate inflammatory stress damage of skin.
4. Barrier repair pathway (centella asiatica extract): Promote migration and proliferation of keratinocytes and fibroblasts to support epidermal repair; downregulate MMP‑1 expression, slow telomere shortening of fibroblasts and boost collagen synthesis.
5. Matrix homeostasis pathway: Upregulate SOD2 and SIRT3 expression to preserve mitochondrial function; raise hydroxyproline level and stimulate collagen production.

Experimental Data

1. Human facial wrinkle test (3% Serum): After 30 minutes, the quantity and severity of facial wrinkles decrease significantly; the count and severity of crow’s feet are markedly improved.
2. Collagen synthesis test: At sample concentration of 20 μg/mL, hydroxyproline (collagen marker) content increases by 106%.
3. Cell scratch migration test: At 250 μM, fibroblast wound healing rate rises by approx. 20%; at 500 μM, keratinocyte wound healing rate rises by approx. 48.6%.
4. Mitochondrial protein expression test: At 10 μM sample, SOD2 expression increases by approx. 40% and SIRT3 expression increases by approx. 75%.
5. Antioxidant test: At 5 mg/mL, DPPH scavenging rate reaches 91% and ABTS⁺ scavenging rate reaches 95%.
